MARIPOSA demonstrates overall survival benefit from amivantamab–lazertinib

The third-generation EGFR tyrosine kinase inhibitor (TKI) osimertinib is the standard-of-care first-line therapy for patients with advanced-stage non-small-cell lung cancer (NSCLC) harbouring EGFR exon 19 deletions or EGFRL858R mutations. Given that 25–40% of these patients die before they can receive second-line therapy, improved first-line options are needed. The phase III MARIPOSA trial tested one potential option by adding the EGFR × MET bispecific monoclonal antibody amivantamab to lazertinib, another third-generation EGFR TKI, with initial results demonstrating a progression-free survival benefit. Now, updated results from MARIPOSA show that amivantamab–lazertinib also improves overall survival (OS).

A total of 1,074 patients were randomly allocated (2:2:1) to receive amivantamab–lazertinib versus monotherapy with either osimertinib or lazertinib. OS, a secondary end point, was immature in a previous report at a median follow-up of 22 months.
